Anti-amoebic

Contraindication

Hypersensitivity to metronidazole and other nitroimidazoles.

Special Precautions

Patient w/ CNS disease, history of blood dyscrasias, ESRD. Re-administer immediately after haemodialysis. Severe hepatic impairment esp hepatic encephalopathy. Pregnancy and lactation. Patient Counselling May impair ability to drive or operate machinery. Monitoring Parameters Monitor total and differential leukocyte counts.

Side Effects

GI disturbances (e.g. nausea, anorexia, unpleasant metallic taste, vomiting, diarrhoea, abdominal discomfort, constipation); furred tongue, glossitis, stomatitis; weakness, dizziness, ataxia, headache, drowsiness, insomnia, hallucinations, changes in mood or mental state (e.g. depression or confusion); urethral discomfort, darkening of the urine; rashes, urticaria, pruritus; raised liver enzyme values, cholestatic hepatitis, jaundice, pancreatitis, thrombophlebitis (IV). Rarely, agranulocytosis, leucopenia, pancytopenia, thrombocytopenia, erythema multiforme, angioedema, anaphylaxis, antibiotic-associated colitis.

Drug Interactions

Concurrent use w/ disulfiram may produce psychotic reactions. May potentiate the effect of oral anticoagulants. May increase risk of lithium toxicity. May reduce the renal clearance resulting to increased toxicity of 5-fluorouracil. May increase serum levels of ciclosporin. May increase plasma levels of busulfan resulting to severe busulfan toxicity. Enhanced metabolism w/ phenobarbital and phenytoin resulting to decreased serum concentrations.
